    Effect of Zn doping on magnetic order and superconductivity in LaFeAsO

    We report Zn-doping effect in the parent and F-doped LaFeAsO oxy-arsenides. Slight Zn doping in LaFe1−x_{1-x}Znx_{x}AsO drastically suppresses the resistivity anomaly around 150 K associated with the antiferromagnetic (AFM) spin density wave (SDW) in the parent compound. The measurements of magnetic susceptibility and thermopower confirm further the effect of Zn doping on AFM order. Meanwhile Zn doping does not affect or even enhances the TcT_c of LaFe1−x_{1-x}Znx_{x}AsO0.9_{0.9}F0.1_{0.1}, in contrast to the effect of Zn doping in high-TcT_c cuprates. We found that the solubility of Zn content (xx) is limited to less than 0.1 in both systems and further Zn doping (i.e., xx ≥\geq 0.1) causes phase separation. Our study clearly indicates that the non-magnetic impurity of Zn2+^{2+} ions doped in the Fe2_2As2_2 layers affects selectively the AFM order, and superconductivity remains robust against the Zn doping in the F-doped superconductors.Comment: 7 figures, 13 pages; revised version with more dat

    Machine Fault Classification Based on Local Discriminant Bases and Locality Preserving Projections

    Machine fault classification is an important task for intelligent identification of the health patterns for a mechanical system being monitored. Effective feature extraction of vibration data is very critical to reliable classification of machine faults with different types and severities. In this paper, a new method is proposed to acquire the sensitive features through a combination of local discriminant bases (LDB) and locality preserving projections (LPP). In the method, the LDB is employed to select the optimal wavelet packet (WP) nodes that exhibit high discrimination from a redundant WP library of wavelet packet transform (WPT). Considering that the obtained discriminatory features on these selected nodes characterize the class pattern in different sensitivity, the LPP is then applied to address mining inherent class pattern feature embedded in the raw features. The proposed feature extraction method combines the merits of LDB and LPP and extracts the inherent pattern structure embedded in the discriminatory feature values of samples in different classes. Therefore, the proposed feature not only considers the discriminatory features themselves but also considers the dynamic sensitive class pattern structure. The effectiveness of the proposed feature is verified by case studies on vibration data-based classification of bearing fault types and severities

    Fault Diagnosis of Motor Bearing by Analyzing a Video Clip

    Conventional bearing fault diagnosis methods require specialized instruments to acquire signals that can reflect the health condition of the bearing. For instance, an accelerometer is used to acquire vibration signals, whereas an encoder is used to measure motor shaft speed. This study proposes a new method for simplifying the instruments for motor bearing fault diagnosis. Specifically, a video clip recording of a running bearing system is captured using a cellphone that is equipped with a camera and a microphone. The recorded video is subsequently analyzed to obtain the instantaneous frequency of rotation (IFR). The instantaneous fault characteristic frequency (IFCF) of the defective bearing is obtained by analyzing the sound signal that is recorded by the microphone. The fault characteristic order is calculated by dividing IFCF by IFR to identify the fault type of the bearing. The effectiveness and robustness of the proposed method are verified by a series of experiments. This study provides a simple, flexible, and effective solution for motor bearing fault diagnosis. Given that the signals are gathered using an affordable and accessible cellphone, the proposed method is proven suitable for diagnosing the health conditions of bearing systems that are located in remote areas where specialized instruments are unavailable or limited

    Development and internal validation of a nine-lncRNA prognostic signature for prediction of overall survival in colorectal cancer patients

    Background Colorectal cancer remains a serious public health problem due to the poor prognosis. In the present study, we attempted to develop and validate a prognostic signature to predict the individual mortality risk in colorectal cancer patients. Materials and Methods The original study datasets were downloaded from The Cancer Genome Atlas database. The present study finally included 424 colorectal cancer patients with wholly gene expression information and overall survival information. Results A nine-lncRNA prognostic signature was built through univariate and multivariate Cox proportional regression model. Time-dependent receiver operating characteristic curves in model cohort demonstrated that the Harrell’s concordance indexes of nine-lncRNA prognostic signature were 0.768 (95% CI [0.717–0.819]), 0.778 (95% CI [0.727–0.829]) and 0.870 (95% CI [0.819–0.921]) for 1-year, 3-year and 5-year overall survival respectively. In validation cohort, the Harrell’s concordance indexes of nine-lncRNA prognostic signature were 0.761 (95% CI [0.710–0.812]), 0.801 (95% CI [0.750–0.852]) and 0.883 (95% CI [0.832–0.934]) for 1-year, 3-year and 5-year overall survival respectively. According to the median of nine-lncRNA prognostic signature score in model cohort, 424 CRC patients could be stratified into high risk group (n = 212) and low risk group (n = 212). Kaplan–Meier survival curves showed that the overall survival rate of high risk group was significantly lower than that of low risk group (P < 0.001). Discussion The present study developed and validated a nine-lncRNA prognostic signature for individual mortality risk assessment in colorectal cancer patients. This nine-lncRNA prognostic signature is helpful to evaluate the individual mortality risk and to improve the decision making of individualized treatments in colorectal cancer patients

    Cdkn2a (Arf) loss drives NF1-associated atypical neurofibroma and malignant transformation

    Plexiform neurofibroma (PN) tumors are a hallmark manifestation of neurofibromatosis type 1 (NF1) that arise in the Schwann cell (SC) lineage. NF1 is a common heritable cancer predisposition syndrome caused by germline mutations in the NF1 tumor suppressor, which encodes a GTPase-activating protein called neurofibromin that negatively regulates Ras proteins. Whereas most PN are clinically indolent, a subset progress to atypical neurofibromatous neoplasms of uncertain biologic potential (ANNUBP) and/or to malignant peripheral nerve sheath tumors (MPNSTs). In small clinical series, loss of 9p21.3, which includes the CDKN2A locus, has been associated with the genesis of ANNUBP. Here we show that the Cdkn2a alternate reading frame (Arf) serves as a gatekeeper tumor suppressor in mice that prevents PN progression by inducing senescence-mediated growth arrest in aberrantly proliferating Nf1−/− SC. Conditional ablation of Nf1 and Arf in the neural crest-derived SC lineage allows escape from senescence, resulting in tumors that accurately phenocopy human ANNUBP and progress to MPNST with high penetrance. This animal model will serve as a platform to study the clonal development of ANNUBP and MPNST and to identify new therapies to treat existing tumors and to prevent disease progression

    Energy deficiency promotes rhythmic foraging behavior by activating neurons in paraventricular hypothalamic nucleus

    BackgroundDysregulation of feeding behavior leads to a variety of pathological manifestations ranging from obesity to anorexia. The foraging behavior of animals affected by food deficiency is not fully understood.MethodsHome-Cage system was used to monitor the behaviors. Immunohistochemical staining was used to monitor the trend of neuronal activity. Chemogenetic approach was used to modify neuronal activity.ResultsWe described here a unique mouse model of foraging behavior and unveiled that food deprivation significantly increases the general activities of mice with a daily rhythmic pattern, particularly foraging behavior. The increased foraging behavior is potentiated by food cues (mouthfeel, odor, size, and shape) and energy deficit, rather than macronutrient protein, carbohydrate, and fat. Notably, energy deficiency increases nocturnal neuronal activity in paraventricular hypothalamic nucleus (PVH), accompanying a similar change in rhythmic foraging behavior. Activating neuronal activity in PVH enhances the amplitude of foraging behavior in mice. Conversely, inactivating neuronal activity in PVH decreases the amplitude of foraging behavior and impairs the rhythm of foraging behavior.DiscussionThese results illustrate that energy status and food cues regulate the rhythmic foraging behavior via PVH neuronal activity. Understanding foraging behavior provides insights into the underlying mechanism of eating-related disorders

    Bidirectional cross-regulation between the eNOS and ß-catenin signalling pathways

    AIMS: β-catenin has been shown to be regulated by inducible nitric oxide synthase (NOS) in endothelial cells. We investigated here whether β-catenin interacts with and regulates endothelial NOS (eNOS) and whether eNOS activation promotes β-catenin signalling. METHODS AND RESULTS: We identified β-catenin as a novel eNOS binding protein in human umbilical vein endothelial cells (HUVECs) by mass spectroscopy and western blot analyses of β-catenin and eNOS immunoprecipitates. This was confirmed by in situ proximity ligation assay. eNOS activity, assessed by cGMP production and eNOS phosphorylation (Ser1177), was enhanced in β-catenin(-/-) mouse pulmonary endothelial cells (MPECs) relative to wild-type MPECs. eNOS activation (using adenosine, salbutamol, thrombin, or histamine), or application of an NO donor (spermine NONOate) or cGMP-analogue (8-bromo-cGMP) caused nuclear translocation of β-catenin in HUVEC as shown by western blotting of nuclear extracts. Exposure to spermine NONOate, 8-bromo-cGMP, or sildenafil (a phosphodiesterase type 5 inhibitor) also increased the expression of β-catenin-dependent transcripts, IL-8, and cyclin D1. Stimulation of wild-type MPECs with basic fibroblast growth factor (bFGF), vascular endothelial growth factor (VEGF), spermine NONOate, 8-bromo-cGMP, or sildenafil increased tube length relative to controls in an angiogenesis assay. These responses were abrogated in β-catenin(-/-) MPECs, with the exception of that to bFGF which is NO-independent. In C57BL/6 mice, subcutaneous VEGF-supplemented Matrigel plugs containing β-catenin(-/-) MPECs exhibited reduced angiogenesis compared with plugs containing wild-type MPECs. Angiogenesis was not altered in bFGF-supplemented Matrigel. CONCLUSION: These data reveal bidirectional cross-talk and regulation between the NO-cGMP and β-catenin signalling pathways

    Epidemiology and cost analysis for patients with oral cancer in a university hospital in China

    <p>Abstract</p> <p>Background</p> <p>Although several studies have reported the direct cost of oral cancer (OC), little research has invested the factors that could influence the costs of OC patient. This study analyzes the epidemiological characteristics and the direct cost of OC. More specifically, the study examines the relationship between patients' medical costs and influencing factors of epidemiology.</p> <p>Methods</p> <p>All patients encountered from January 2007 to December 2007 at the School of Stomatology of the Fourth Military Medical University (FMMU) in China with diagnosis of oral cancer have been selected. Medical hospitalization days (MHD) and cost per patient (CPP) of the samples have been calculated for different patient groups, and the results have been compared using statistical methods.</p> <p>Results</p> <p>A total of 456 oral cancer patients have been selected in this study. The epidemical characteristics are as follows: female/male 176/280; squamous cell carcinoma (SCC)/adenocarcinoma/sarcoma/lymphoma/other types 246/127/40/27/16; stage I/II/III/IV 90/148/103/115; smoker/non-smoker 136/320; rural/urban patients 82/374. Of all the patients, 82.24% were over 40 years of age. Rural patients were significantly younger than urban patients. SCC was the majority histology in older patients, while sarcoma was more common in younger patients. 372 of the patients received treatment and 84 gave up any treatment after diagnosis. Treatment cost accounted for majority of the payment. The CPP and MHD of patients in late clinical stage were higher than that of patient in early stage.</p> <p>Conclusion</p> <p>Gender, smoking habit and age older than 40 years are the epidemiological risk factors for oral cancer. Lack of medicare, smoking habit, late clinical stage and SCC are the high economic factors for patient medical cost.</p
